Angiogenesis, the formation of new vessels from a preexisting vasculature depends critically on the ability of the capillary endothelial cells to sever their normal cellcell associations, to migrate through extracellular matrix, and to reform their cellcell attachments to generate new capillaries. In the initial stages, adhesive interactions between endothelial cells and the extracellular matrix are dominant thus facilitating endothelial cell invasion of and migration through the extravascular matrix. Later, adhesive interactions between endothelial cells are supreme thereby promoting the organization of vascular tubes. The adhesive interactions of endothelial cells with each other and with the extracellular matrix, along with the adhesion receptors that mediate these interactions, are thus of fundamental importance to this process.1,2 This indicates important roles for endothelial cellcell adhesion molecules and endothelial cellmatrix adhesion molecules (e.g., integrins) in the formation of new vessels. The participation avb3 and avb5 integrins in in vivo angiogenesis has been demonstrated and progress has made in understanding their role in vessel formation (discussed in Chapter 3). Recently, evidence has emerged to implicate another endothelial cellcell adhesion molecule, PECAM1, in angiogenesis.3 In this chapter we will review the data that has established a role for PECAM1 in in vivo angiogenesis as well as the emerging data which suggests that it may be involved at several of the stages of the formation of new vessels.
